Research Synopsis
In my PhD, I develop a novel high-throughput platform (uASPIre) for sequence-function mapping of gene regulatory elements in Escherichia coli. I combine a recombinase-based DNA recorder with next-generation sequencing, which enables us to record sequence-function data of millions of genetic variants with high phenotypic and temporal resolution at low experimental effort. I capitalize on these features to develop an in-depth understanding of key sequence determinants governing translation in bacteria.
